چکیده
Background: MicroRNA (miR) has been suggested in the development of several types cancer; yet, exact function miR-4284 colon cancer remains elusive.
Methods: MiR-4284 expression was assessed normal cell line CCD-18Co, and HT-29 SW480 lines representing human cancer. Potential target gene predicted using TargetScanHuman, experimentally verified luciferase report assay. Wound-healing, invasion attachment were evaluated to determine effect on migration, invasion, metastatic properties lines. Expression epithelial-mesenchymal transition (EMT) phenotypic protein hallmarks, including N-cadherin, E-cadherin, as well Vimentin, also evaluated.
Results: significantly decreased lines, Perilipin 5 (PLIN5) found be directly targeted by miR-4284. Ectopic reduced endogenous level PLIN5 suppressing phenotypes. In addition, re-introducing reversed profile EMT markers.
Conclusion: Our findings for first time identify an anti-tumor miRNA cancer, which acts reduce inhibit EMT, leading inhibited tumorigenesis. These results highlight potential a therapeutic

Ginsenoside Rb1 inhibits hypoxia-induced epithelial-mesenchymal transition in ovarian cancer cells by regulating microRNA-25
Metastasis frequently occurs in advanced ovarian cancer, which not only leads to substantial mortality but also becomes a major challenge to effective treatment. Epithelial-mesenchymal transition (EMT) is a key mechanism facilitating cancer metastasis. Targeting the EMT process with more efficacious and less toxic agents to prevent metastasis is of significant therapeutic value for ovarian canc...
متن کاملGinsenoside Rg3 inhibits epithelial-mesenchymal transition (EMT) and invasion of lung cancer by down-regulating FUT4.
The epithelial-mesenchymal transition (EMT) is an important factor in lung cancer metastasis, and targeting EMT is a potential therapeutic strategy. Fucosyltransferase IV (FUT4) and its synthetic cancer sugar antigen Lewis Y (LeY) was abnormally elevated in many cancers. In this study, a traditional Chinese medicine ginsenoside Rg3 was used to investigate whether its inhibition to EMT and invas...
متن کاملMicroRNA-200c overexpression inhibits tumorigenicity and metastasis of CD117+CD44+ ovarian cancer stem cells by regulating epithelial-mesenchymal transition
BACKGROUND Cancer stem cells (CSCs) are believed to be 'seed cell' in cancer recurrence and metastasis. MicroRNAs (miRNAs) can play an important role in the progression of primary tumor towards metastasis by regulating the epithelial-mesenchymal transition (EMT). The goal of this study was to investigate the effect of miRNA-200c overexpression on the EMT, tumorigenicity and metastasis of epithe...
متن کاملMicroRNA-224 inhibits proliferation and migration of breast cancer cells by down-regulating fizzled 5 expression
The Wnt/β-catenin signaling is crucial for the proliferation and migration of breast cancer cells. However, the expression of microRNA-224 (miR-224) in the different types of breast cancers and its role in the Wnt/β-catenin signaling and the proliferation and migration of breast cancer cells are poorly understood. In this study, the levels of miR-224 in different types of breast cancer tissues ...
متن کاملmiR-573 inhibits prostate cancer metastasis by regulating epithelial-mesenchymal transition
The metastastic cascade is a complex process that is regulated at multiple levels in prostate cancer (PCa). Recent evidence suggests that microRNAs (miRNAs) are involved in PCa metastasis and hold great promise as therapeutic targets. In this study, we found that miR-573 expression is significantly lower in metastatic tissues than matched primary PCa. Its downregulation is correlated with high ...
